Take a look at the size of the 3D Manipulator Handles in the 3D window as you change these settings. You should compare this to playing with the DPI setting itself, but I find that doubling the virtual pixel setting instead of the DPI (or in addition to DPI) provides a more natural look, particularly in the 3D window itself. This has the effect of making small pixel-based graphics much more visible on a high-dpi screen like the Surface Pro/Book. This is right under the DPI setting in User Preferences -> System. This may just be personal preference on my part, but I strongly prefer the non-emulated button on a real mouse. Blender does have a mode where it can emulate that for use with a touchpad. I like using the real middle mouse button. Performance seems great, but I have two recommendations for the interface: I installed Blender on a Surface Pro 4, which has a very similar screen but a somewhat less powerful GPU than the Surface Book.
